Yes, your train needs to arrive before midnight in Hiroshima. Minimum 
journey time from Tokyo station is a little under 4 hours (3:47). 
The hyperdia link will show you options an connection time, best
place to do that is Shinagawa, if you decide to do it in Tokyo
give yourself some extra time, it is a BIG station.

Yes, you can buy beer on the train. If you are in "Green Car" (first 
class) they bring around a cart, otherwise every train has a dining
car and I think there are even vending machines on some trains.
